New Test Method for the measurement of total bacteria in fuel and fuel associated water using quantitative polymerase chain reaction (qPCR)
1. Scope
This test method covers a protocol for capturing, preserving and purifying nucleic acids and quantifying total bacterial content in fuel and fuel-associated water using quantitative polymerase chain reaction.
There is no standard for qPCR in fuel - several years ago a standard practice was published but lacks specific methodological details. qPCR is a more commonly used method for microbial detection and quantification and the industry would benefit from a standard method.
