New Test Methods for Nucleic Acid Extraction and Quantitative PCR analysis in Environmental Water Samples

1. Scope

This Test Method discusses DNA extraction of environmental samples from environmental waters. It also discusses pre-processing of samples, removal of inhibitors, cleanup procedures, quality assurance procedures, data qualifiers, and corrective actions. This Test Method does discuss the subsequent use of extracted nucleic acids in quantitative polymerase chain reaction.

Keywords

Dehalococcoides; groundwater; bioremediation; monitored natural attenuation (MNA);

Rationale

The groundwater community needs a test method for the procedures used to assess biodegradation activity in the subsurface so that users can know that the results provided by one laboratory are comparable to those developed by another laboratory. Consistency in procedures and results will foster greater usage of these molecular tools.

The title and scope are in draft form and are under development within this ASTM Committee.
