Polarographic Measurement of Dissolved Oxygen

Polarographic techniques have been found to be generally applicable for the determination of dissolved oxygen. The operating voltage for any system is selected from a current-voltage curve. For most systems commonly encountered, 0.6v can be used. At the operating voltage the current is directly proportional to the oxygen content. Residual current is determined following a nitrogen purge of the solution. Calibration is accomplished by preparing a curve of diffusion current versus dissolved oxygen using a modified Winkler technique as a standard. The calibration can be checked on air saturated solutions whose dissolved oxygen content is known from handbook solubilities. Modified sampling and analytical techniques are described for dissolved oxygen and oxygen utilization measurements in the presence of biological sludges. A comparison of the polarographic method with chemical methods for several substances is presented. The effects of the impurities of peptone and castile soap on the oxygen polarogram and the polarographic method are discussed.
