Electron Probe X-ray Microanalysis of Medical and Biological Specimens

The need for local microchemical analysis at the micron level and a summarized review of the literature on the electron probe application in biology and medicine are presented. The author's procedures for preparation of biological specimens, electron probe effects on the specimen, and results from microanalysis studies on the following topics are given: (1) calcified corpuscles used as standard test specimens, (2) normal growth of the mammalian long bone, (3) hardening of blood vessels, (4) degeneration of cancer cells, and (5) Wilson's disease. Qualitative data are presented in all cases, and quantitative data for calcium and phosphorus in both normal and pathologic calcification processes. Light microscopy, electron microscopy, X-ray contact microradiography, and X-ray diffraction were carried out simultaneously. Some of these results are correlated with those obtained by the electron probe microanalysis method.
