A Technique for Easy Removal of Direct Replicas for Electron Microscopy

Source

The advantages of shadowing directly on the specimen surface in the preparation of replicas for electron microscopy are twofold. First, there is no ambiguity to the interpretation-one does not have to invert the shadow direction. Second, there is no chance for artifacts due to swelling, shrinking, or straining such as are apt to develop with replicas shadowed after removal. There is, however, a serious difficulty which arises when shadowing a metal directly on a clean metal surface. The replicas are invariably bound so tightly to the specimen surface that it is necessary to etch away the specimen to release the replica. The chemical attack usually results in breaking the replica into small pieces and negates the re-replication of the specimen without repolishing and etching.
